Daisy von Scherler Mayer was born into a Hollywood family - her grandfather was the famed Hollywood screenwriter Edwin Justus Mayer; her mother was actress Sasha von Scherler and her father, writer Paul Avila Mayer, created the soap opera Ryan's Hope.

Von Scherler Mayer worked behind the scenes at the New York Shakespeare Festival when she was a teenager, and later graduated with a degree in theater and history from Wesleyan University. In 1995 she directed and co-wrote the feature Party Girl, starring Parker Posey. The film was nominated for the Grand Jury Prize at the Sundance Festival, and was such a success that FOX television bought the rights to turn it into a television series in 1996. Von Scherler Mayer next directed the feature Woo (1998), starring Jada Pinkett Smith, and although that film bombed both at the box office and with critics, von Scherler Mayer was busy directing her next film, Madeline (1998), based on Ludwig Bemelman's classic children's books about the little red-haired French girl. The movie was a hit with audiences and critics alike. Von Scherler Mayer waited for the right project to come along before deciding to direct The Guru (2003), a comedy starring Heather Graham, Marisa Tomei and newcomer Jimi Mistry.

Von Scherler Mayer currently lives in Brooklyn, New York with her husband.
