Born in New York City, Nicole Holofcener studied film making at Columbia Graduate School, where she wrote and directed the short film It's Richard I Love. Upon graduating, she wrote the screenplay for Walking and Talking, bought by Good Machine studios.

Holofcener decided to expand her knowledge of film and attended a workshop at the writers' and then the directors' lab at the Sundance Institute. Around the same time, Good Machine filmed Holofcener's short Angry, which she both wrote and directed. In 1996 Walking And Talking was released to theater and film festivals where it earned a nomination for Grand Special Prize from the Deauville Film Festival.

Aside from feature films, Holofcener has also written many scripts for television studios, and directed a number assignments, including episodes of Gilmore Girls and Six Feet Under.

In 2002 she released her second film, Lovely & Amazing, that she again wrote and directed. About a family that adopts an African-American child, the story somewhat reflects Hologcener's own family, as she has an adoptive African-American brother.

For her third film, Friends with Money, Holofcener received the Dorothy Arzner Directors Award.

Married, Holofcener has two children with her husband, Benjamin Allanoff.
